Alfonso was born in 1275 in Great Hormead, Royston, Hertfordshire, England.1 Alfonso's father was Sir Robert de Vere Earl of Oxford, Duke of Ireland and his mother was Alice de Sanford. His paternal grandparents were Hugh de Vere and Hawise de Quincy; his maternal grandparents were Gilbert de Sanford and Loretta la Zouche. He had four brothers and three sisters, named Robert, Gilbert, Philip, Hugh, Joan, Hawise and Lora.

John de Vere was the only son of Alfonso de Vere, and Jane, daughter of Sir Richard Foliot. Alfonso was a younger son of Robert de Vere, 5th Earl of Oxford, and brother of Robert de Vere, 6th Earl of Oxford. When the younger Earl Robert's son died without issue in 1329, the earl obtained licence from the king to entail his estates on his nephew, John.
